LAB PURPOSE
In the broadest sense, this exercise is designed to provide students with hands-on experience in enzyme chemistry.  More specifically, the hydrolysis of starch by a-amylase is used as an example to illustrate several key concepts:
  • the importance of consistency in experimental technique;
  • the necessity of producing standard curves and zeroing blanks in order to quantify data;
  • how the maximum rate of an enzyme can be determined;
  • the effect of enzyme and substrate as limiting factors; and
  • how and why pH and temperature affect the function of an enzyme.
